Abstract

Tujuan dari penelitian ini adalah untuk menumbuhkan kultur murni mikroorganisme dan meneliti sifat-sifat morfologis dan fisiologisnya dengan menggunakan media agar padat dan cair, teknik isolasi dan pengenceran progresif digunakan untuk membuat kultur murni. Koloni yang menunjukkan ciri-ciri yang berbeda dipilih untuk dibiakkan kembali setelah diisolasi dari sampel lingkungan yang mengandung bakteri. Pewarnaan gram, bentuk sel, morfologi koloni, dan uji biokimia untuk menentukan kapasitas metabolisme mikroorganisme adalah beberapa sifat mikrobiologis yang diamati. Temuan menunjukkan bahwa mikroorganisme yang berhasil diisolasi berbeda dalam hal struktur koloni, pewarnaan, dan hasil uji biokimia yang mengungkapkan aktivitas fermentasi tertentu. Menurut perdebatan tersebut, kultur murni yang representatif untuk investigasi tambahan dapat dihasilkan dengan menggunakan teknik isolasi yang tepat. Kultur murni dapat dibuat dengan menggunakan teknik isolasi yang tepat, dan identifikasi spesies mikroba serta kemungkinan penggunaannya di berbagai bidang dapat dibantu dengan pengamatan sifat-sifat mikroba.
